Resumen

Genome resource banks are the base for domestic and wild species genetic selection, so it is important to focus in achieving the highest quality on the samples we preserve. Although that is not always possible we have alternatives to improve post-thawing semen quality such as sperm selection gradients. They aim to minimize the damage that cryopreservation causes in the sperm cells. In this study we have tested the Androcoll single layer centrifugation gradient with the objective of observing its utility on post-thaw semen samples of ram and red deer within an hour of incubation at 37 °C. We have also tested the presence of mitochondrial superoxide with the fluorescent probe MitoSOX. We use a pool of 3 males from each species. The gradient increased significantly the quality of the post thawed samples, obtaining a higher number of active mitochondria on the selected population compared to the control. Regarding the incubation we found differences between the species. For red deer samples we found a significant improvement in viability and active mitochondria results that we did not found with the ram samples after 1 hour of incubation.
