Resumo

The application of reproductive techniques is important for the conservation and farming of amphibians. Studies on the status of sperm chromatin are lacking on this group. Therefore, we have tested the sperm chromatin structure assay (SCSA) on 14 males of Bufocalamita spermatozoa. Spermiation was induced by hCG, and spermatozoa were analyzed in two successive collections separated by 2 h. DNA fragmentation (%DFI) was low or moderate, below cut-off values for mammals (30%), with males showing a great heterogeneity. Moderate values decreased in the second collection. Chromatin compaction (%HDS) was more homogeneous between males and mostly below 10%. A principal components analysis with SD-DFI, %DFI and %HDS at both collection times allowed to classify the males according their sperm chromatin characteristics and changes between recovery times. Moreover, fragmentation (SD-DFI and %DFI) and compaction (%HDS) were largely independent variables. SCSA could be useful for studying amphibian sperm chromatin for different purposes and regarding the application of reproductive techniques.
